The rapper was joined by his wife Beyonce and an exclusive guest list at his New York nightspot and no expense was spared to mark the special occasion.
Revellers enjoyed Ace of Spades champagne and D'usse shots served by waitresses wearing dresses adorned with 14-carat gold stitching.
The packed VIP lounge included Kylie Minogue, Victor Cruz, Ray Kelly and recently divorced diet specialist Bethenny Frankel.
According to the New York Post newspaper, the ringed off section was so busy at one point pop princess Kylie couldn't get out.
After struggling to leave for 10 minutes, the singer finally freed herself, prompting Jay-Z to tell security the section was ''closed''.
Also in attendance was NFL mogul Robert Kraft and his girlfriend Ricki Noel Lander, and the New England Patriots team owner shared a joke with Jay, about his Super Bowl ring - which he recently accused Russian leader Vladimir Putin of stealing.
An onlooker said: ''Jay touched Kraft's championship ring, and Kraft asked if he wanted it. Jay said, 'I'm going to take it!' ''
Jay then joked to friends: ''I just bought the New England Patriots.''
The 40/40 club is named after a baseball term for the elite club of players who have hit 40 home runs and stolen 40 bases in one season. There are only four players in history to complete this feat; Jose Canseco, Alex Rodriguez, Barry Bonds and Alfonso Soriano.
